Tomography studies of biological cells on polymer scaffolds

Abstract

Cells seeded on polymer yarns, stained and measured with synchrotron radiation micro-computed tomography (SRμCT) in absorption and in differential absorption contrast mode, were studied. A new cell segmentation procedure was devloped and the measured volumetric data were quantitatively assessed. Quantification parameters included total cell volume, cell volume density, total yarn volume, and the radial cell mass distribution. It was shown that the applied variation of the staining parameter of gold enhancement incubation time had significant influence on the cell volume density. It was found that differential absorption contrast mode provided similar but no additional.
