Regulation of amyloid precursor protein processing by lithium

Abstract

Secreted A β is produced during normal cell metabolism and it is regulated by many factors. Quantification of A β generation is an important approach to studying APP processing. In this chapter, we demonstrate that lithium, a potent GSK-3 inhibitor, can regulate amyloid secretion in COS7 cells overexpressing APPC100 demonstrated by a sandwich enzyme-linked immunosorbent assay (ELISA). Finally, we show that lithium can effectively inhibit tau pathology induced by A β injection into the hippocampus of a transgenic mouse model.
