Multicolumn high resolution LC-MALDI MS system for high throughput analysis

Abstract

A multicolumn high resolution LC-MALDI MS system was developed for high throughput proteomic analysis. Complex samples were fractionated by strong cation exchange chromatography into 20 or more fractions. The UltiMate nano-LC system was optimized to operate in the automatic multicolumn nano-LC mode. The use of peptide trap precolumns allowed preconcentration of diluted samples and rapid loading of the sample at flow rate of up to 5 μl/min. The MS spectra were processed using cross-correlation algorithm and m/z values corresponding to peaks with chromatographic behavior were selected for peptide mass fingerprinting and/or MS/MS interrogation.
