Abstract
An algorithm on the fundamental advantages of LC-MS involving high-resolution separation and archiving of data was discussed. A Peak-picking software based on the algorithm was developed. The software was successfully tested in the analysis of a tryptic digest of a mixture of 9 proteins. The advantage of this correlation function approach lie on it's ability to calculate more precisely peak centroids and quantitate the amount of corresponding peptide by using a priori information about the shapes of chromatographic and MS peaks. The algorithm can be used for LC LC(CE)-MALDI-MS in addition to LC (CE)-ESI-MS data.
