The synthesis and LH releasing activity of [8 homoarginine] LH RH are described. Its biological activity is discussed in relation to that of other analogues with substitutions in position 8. Although in homoarginine the positive charge of the guanidino group of arginine is only displaced by 1.54 A, the LH releasing activity is reduced to 20% of that of LH RH. There is a surprising analogy with ACTH. Replacing 8 arginine by Å ornithine, or homoarginine, nearly the same decrease in the biological activity has been found as with the correpsonding analogues of LH RH.
